Her smiling at you multiple times means she doesn't find you creepy, and would likely be receptive to a conversation. It doesn't necessarily means she wants to fuck you, but if you play your cards right, she will.

Just say something about a shirt she is wearing (like if it has something on it), or make conversation about the gym. Ask her how's her lifts are doing or something. When she smiles, is she showing her teeth?

Her friend is intimidating, sure, but she probably won't interfere. You're gonna have to read the situation if she wants to continue talking or not. Be relaxed, and confident. When you get a little conversation going, ask for her number, and excuse yourself back to your workout. Don't overstay your welcome, so keep it no longer than a few minutes, and try to be self-aware of how you may be coming off (annoying, creepy, welcomed, or whatever). I know this is easier said than done, trust me, I know, but you gotta approach if you want some.
